As you can tell I’ve been doing a bit of SEO work today and customising my Blog. It’s made me think - should I be using American English rather than British English on my blog to improve my Search Engine Optimisation/Optimization?

It depends on what you want to achieve with ranking and what visitors will think of reading real English versus American. For my audience real English is a positive, therefore I wouldn't want to devalue myself by sounding like an American even if it improved ranking. Content and quality is still king.

Given that this is a UK blog I've resisted using American English. Yes, I want more traffic but at the moment I'm not that desperate (yet) for traffic.
